What makes Paper 4 different
If you are sitting IGCSE Biology Paper 4, you are an Extended candidate. That means you have covered everything in the Core syllabus plus the Supplement content, and you are aiming for grades A* to E. Paper 4 is a written theory paper lasting 1 hour and 15 minutes, worth 80 marks. Unlike the multiple-choice papers, every mark here depends on what you write, how precisely you write it, and whether you answer the question that was actually asked.
Think of Paper 4 like a conversation with the examiner, except you only get one chance to say the right things. The examiner cannot ask you to clarify. They read your answer, check it against the mark scheme, and award marks for every correct point you make. Your job is to make those points clearly, using the right scientific language, in the right amount of detail.
The paper structure at a glance
Paper 4 is made up of structured questions, which means each question is broken into parts: (a), (b), (c), and sometimes further into (i), (ii), (iii). The marks for each part are printed in brackets at the end of the line. This is your single most important navigation tool.

The key difference is not just the content but the depth. Paper 4 expects you to explain mechanisms, not just name them. Knowing that "osmosis is the movement of water" is enough for Core. For Extended, you need to say it is the net movement of water molecules through a partially permeable membrane from a region of higher water potential to a region of lower water potential.
Time management: one mark, one minute
With 80 marks and 75 minutes, you have just under one minute per mark. That is a useful rule of thumb, but not every mark takes the same effort. A one-mark "State" question might take 15 seconds. A six-mark "Explain" question might take seven or eight minutes because you need to build a logical chain.
A common mistake is spending too long perfecting early answers and then rushing through the final questions. The last questions on the paper are often worth the most marks, and they tend to test the harder Supplement content. Running out of time there costs you disproportionately.
Command words: the examiner's instructions
Every IGCSE Biology question begins with a command word that tells you exactly what kind of answer is expected. Think of command words as a recipe: they tell you the ingredients your answer needs. The difference between "Describe" and "Explain" catches more students than any other distinction. If a question says "Describe the effect of increasing temperature on enzyme activity," you need to say what happens: activity increases, reaches an optimum, then decreases. If it says "Explain," you also need to say why: more kinetic energy means more frequent collisions between enzyme and substrate, but beyond the optimum the active site shape changes due to denaturation, so the substrate no longer fits.
How to tackle high-mark questions
Paper 4 regularly includes questions worth five, six, or even seven marks. These are where the strongest candidates pull ahead. The secret is structure: examiners award marks for individual points, not for overall impression. A beautifully written paragraph that only contains two marking points scores two marks, regardless of how elegant it sounds.
The one-point-per-line approach
For a six-mark question, aim to make at least seven or eight distinct points. Write each one as a separate sentence or on a new line. This forces clarity and makes it easy for the examiner to spot your marking points. Think of it like laying out your evidence on a table rather than bundling it all into a bag.
Build a logical chain
Many high-mark questions on Paper 4 ask you to explain a process or mechanism. The marks often follow a logical sequence, and missing a step in the middle can cost you the marks that come after it. For example, if the question asks you to explain why wilting occurs when a plant loses too much water:
- Water is lost from the leaves by transpiration through the stomata.
- If water loss exceeds water uptake from the roots, cells lose water by osmosis.
- The vacuole shrinks and the cytoplasm pulls away from the cell wall.
- This is called plasmolysis (in individual cells) or incipient plasmolysis.
- The cells become flaccid and lose turgor pressure.
- Without turgor pressure to support the stem and leaves, the plant wilts.
Each numbered point above would likely earn a mark. Skip point 3, and the examiner might not award point 4 either, because the chain is broken. Practise these logical sequences when you revise, not just the isolated facts.
Reading mark schemes like an examiner
Cambridge publishes mark schemes for all past papers, and reading them is genuinely one of the best revision techniques for Paper 4. Mark schemes reveal the exact language examiners are looking for. You will notice patterns.
- "Idea of..." means the examiner will accept different wordings, as long as the core concept is there. "Idea of increased surface area" could be awarded for "more surface for absorption" or "larger area for reactions."
- "ORA" stands for "or reverse argument." If the mark scheme says "higher temperature increases rate of reaction (ORA)," you also get the mark for saying "lower temperature decreases rate of reaction."
- Underlined words in mark schemes are essential. If "partially permeable membrane" is underlined, writing "membrane" alone does not score. You need the full term.
- "Max 3" means the question lists four or five possible marking points, but the examiner will award a maximum of three marks. This is good news: you have more ways to earn the marks than you need, so even if you miss one point, you can still get full marks from the others.
Supplement topics that come up repeatedly
Paper 4 tests the full syllabus, but certain Supplement topics appear with striking regularity. These are worth extra revision time because they are almost guaranteed to appear in some form:
- Osmosis and water potential - explaining why water moves in a particular direction, turgor and plasmolysis
- Enzyme kinetics - explaining (not just describing) the effect of temperature and pH, including the mechanism of denaturation
- The nephron - ultrafiltration at the glomerulus, selective reabsorption, role of ADH
- Synapses - chemical transmission across the synaptic cleft
- DNA structure and protein synthesis - base pairing, transcription and translation concepts
- Genetic modification - restriction enzymes, plasmid vectors, transformation of bacteria
- Natural selection leading to evolution - the full logical chain from variation through selection pressure to allele frequency change
- Limiting factors in photosynthesis - interpreting graphs where one factor limits the rate while others do not
If you can write a confident, detailed explanation of each of these topics from memory, you are well prepared for the hardest parts of Paper 4.
Handling unfamiliar context questions
Some of the most challenging questions on Paper 4 present a scenario you have never seen before and ask you to apply your biological knowledge to it. These "Suggest" questions might describe an experiment you have not studied, an organism you have never heard of, or a medical condition that is not in the syllabus.
The trick is to remember that the biology behind these questions is always from the syllabus. The context is new, but the science is not. When you see an unfamiliar question, ask yourself three things:
- Which syllabus topic does this relate to? (Look for clues: if it mentions enzymes, think about active sites, temperature, pH, denaturation.)
- What biological principle applies here? (Diffusion, osmosis, natural selection, negative feedback - the core mechanisms are usually the key.)
- What would the examiner expect me to connect? (The question gives you a scenario. Your job is to bridge from the scenario to the biology using precise language.)
Diagrams and drawings
Paper 4 sometimes asks you to draw or label biological diagrams. These carry easy marks, but only if you follow the conventions:
- Always use a sharp pencil, never a pen, for biological drawings.
- Draw large and clear. A tiny diagram crammed into a corner cannot show the detail examiners need to see.
- Label lines must be straight (use a ruler) and must touch the structure they identify, not just point vaguely near it.
- Do not shade or cross-hatch biological drawings unless specifically instructed.
- If asked to draw a graph, label both axes with the variable name and units. Plot points with small, precise crosses (not dots or circles). Draw a smooth curve or ruled line of best fit as appropriate.
Common mistakes that cost marks on Paper 4
- Writing "Describe" answers for "Explain" questions. If the question says "Explain," you must give reasons, not just a sequence of events. Leaving out the "why" loses you half the available marks.
- Not matching the number of points to the marks available. A three-mark question needs at least three distinct points. Writing one long sentence that contains only one idea scores one mark, no matter how detailed that sentence is.
- Using vague language. "The stuff moves across" does not score. "Glucose is reabsorbed into the blood by active transport in the proximal convoluted tubule" does. Precision wins marks.
- Confusing similar terms. Excretion is not egestion. Respiration is not breathing. Antibodies are not antibiotics. The IGCSE Biology examiners' reports flag these confusions every single year.
- Ignoring data in the question. If a question includes a table or graph, your answer must reference it. Examiners want you to quote specific figures: "At 35 degrees Celsius the rate was 12 cm3 per minute" is far stronger than "the rate was highest at a warm temperature."
- Skipping "Compare" requirements. "Compare" means you must mention both items being compared, not just describe one. "Arteries have thick muscular walls, while veins have thinner walls with valves" is a comparison. "Arteries have thick muscular walls" is only a description of arteries.
- Running out of time on the last questions. The final questions are often the highest-value ones, testing the most demanding Supplement content. Losing marks there because you spent too long on early questions is one of the most avoidable errors.
Using past papers strategically
Past papers are your best revision tool for Paper 4, but the way you use them matters more than how many you complete. Here is a revision approach that builds real exam skill:

- Practise writing under time pressure. Pick three or four high-mark questions from different past papers and answer them in exam conditions. The goal is not to learn new biology but to practise structuring clear, complete answers quickly.
- Read through key definitions and equations. The balanced equations for photosynthesis and respiration, the definition of osmosis, the steps of genetic modification: these are precision-recall facts that you either produce correctly or you do not. A 15-minute review the night before can lock them in.
Walk into the exam hall knowing that you have prepared thoroughly, that you understand the format, and that you have a clear strategy for managing your time. Paper 4 rewards students who combine solid biological knowledge with disciplined exam technique, and both of those are within your control.
